Cytogenetics involves the examination of chromosomes to identify structural  and numerical abnormalities. Chromosomes of a dividing human cell can be analyzed clearly in white blood cells, specifically T lymphocytes, which are easily collected from blood. Cells from other tissues such as bone marrow, amniotic fluid, and other tissues can also be cultured for cytogenetic analysis. Following several days of cell culture, chromosomes arrested at Metaphase stage and are fixed, spread on microscope slides, and stained (GTG-Banding). The staining method allow each of the chromosomes to be individually identified. The distinct bands on each chromosome allows in analysing chromosomal structure.

Cloning and over expression of genes in Arabidopsis and to study the genotypic characters of the transformed plants under different stress conditions.

                      Cloning  of  genes through GATEWAY CLONING TECHNOLOGY and  then mobilizing them in Arabidopsis   plants  by  Agrobacterium mediated transformation   by    Floral Dipping method. Agrobacterium helps in moving  the  gene  of  interest  into  Arabidopsis plant  where the expression of gene allows us to know the effect of  gene upon over expression. These  are further evaluated under different stress conditions to observe the gene response.

 EXTRACTION AND PURIFICATION OF CYCLOTIDES FROM    Tridax procumbens

Cyclotides are small cyclic plant proteins with unique cyclized cystine knot (CCK), which enables stable loop formation with increase stability to with stand thermal, chemical and enzymatic degradations. They show a wide range of biological activities like antimicrobial, anti viral, cytotoxic, insecticidal and immune suppressive activity. These characters make them an ideal tool in pharmaceutical drug applications.

        Extraction begins with the harvesting of  plant  materials and using  solvent  systems total proteins  are  collected, then  the total proteins will be analyzed using  FPLC. As the cyclotides exhibit  hydrophobicity  they  elute  in  the retention time  between   20-30 minutes   (i.e. 20-30 retention time eluents will show highest hydrophobicity). The eluted  peaks will be  collected, pooled and further analyzed by SDS-PAGE  to  identify  the  number  of  hydrophobic proteins (10-20 KD size) resent in  fractions  and  then  their  anti-microbial  activity  is  checked  with different  anti-microbial techniques.
